[eng] Time-of-Flight Positron Emission Tomography (TOF-PET) scanners demand fast and efficient photo-sensors and scintillators coupled to fast readout electronics. Nowadays, there are two main configurations regarding the scintillator crystal geometry: the segmented or pixelated and the monolithic approach. Depending on the cost, spatial resolution and time requirements of the PET module, one can choose between one or another. The pixelated crystal is the most extensive configuration on TOF-PET scanners as the coincidence time resolution is better compared to the monolithic. On the contrary, monolithic scintillator crystals for Time-of-Flight Positron Emission Tomography (ToF-PET) are increasing in popularity this last years due to their performance potential and price in front of the commonly used segmented crystals. On one hand, monolithic blocks allows to determine 3D information of the gamma-ray interaction inside the crystal, which enables the possibility to correct the parallax error (radial astigmatism) at off-center positions within a PET scanner, resulting in an improvement of the spatial resolution of the device. On the other hand, due to the simplicity during the crystal manufacturing process as well as for the detector design, the price is reduced compared to a regular pixelated detector. The thesis starts with the use of HRFlexToT, an ASIC developed in this group, as the readout electronics for measurements with single pixelated crystals coupled to different SiPMs. These measurements show an energy linearity error of 3% and an energy resolution below 10% of the 511 keV photopeak. Single Photon Time Resolution (SPTR) measurements performed using an FBK SiPM NUV-HD (4 mm x 4 mm pixel size) and a Hamamatsu SiPM S13360-3050CS gave a 141 ps and 167 ps FWHM respectively. Coincidence Time Resolution (CTR) measurements with small cross-section pixelated crystals (LFS crystal, 3 m x 3 mm x 20 mm ) coupled to a single Hamamatsu SiPM S13360-3050CS provides a CTR of 180 ps FWHM. Shorter crystals (LSO:Ce Ca 0.4%) coupled to a Hamamatsu S13360-3050CS SiPM or FBK-NUVHD yields a CTR of 117 ps and 119 ps respectively. Then, the results with different monolithic crystals and SiPM sensors using HRFlexToT ASIC will be presented. A Lutetium Fine Silicate (LFS) of 25 mm x 25 mm x 20 mm, a small LSO:Ce Ca 0.2% of 8 mm x 8 mm x 5 mm and a Lutetium-Yttrium Oxyorthosilicate (LYSO) of 25 mm x 25 mm x 10 mm has been experimentally tested. After subtracting the TDC contribution (82 ps FWHM), a coincidence time resolution of 244 ps FWHM for the small LFS crystal and 333 ps FWHM for the largest LFS one is reported. Additionally, a novel time calibration correction method for CTR improvement that involves a pico-second pulsed laser will be detailed. In the last part of the dissertation, a new developed simulation framework that will enable the cross-optimization of the whole PET system will be explained. It takes into consideration the photon physics interaction in the scintillator crystal, the sensor response (sensor size, pixel pitch, dead area, capacitance) and the readout electronics behavior (input impedance, noise, bandwidth, summation). This framework has allowed us to study a new promising approach that will help reducing the CTR parameter by segmenting a large area SiPM into "m" smaller SiPMs and then summing them to recover all the signal spread along these smaller sensors.

Promiscuous signaling of ligands via mutant ALK2 in fibrodysplasia ossificans progressiva

    Get PDF
Fibrodysplasia ossificans progressiva (FOP) is a rare hereditary disorder characterized by successive heterotopic bone formation, for which at present there is no therapy. Mutations in the bone morphogenetic protein (BMP) type I receptor Activin receptor-like kinase 2 (ACVR1/ALK2) are the main trigger for FOP and inflammation is thought to be the secondary hit. The single nucleotide mutation at position 617 in the cDNA ALK2 sequence, which is found in 98% of FOP patients, results in a R206H change in the intracellular juxtamembrane region of ALK2. Previous studies had revealed that this mutation perturbs the interaction with the negative regulator FKBP12, thereby sensitising cells expressing this mutant receptor to BMPs, which are potent inducers of cartilage and bone formation. Recently, however, a twist in the underlying mechanism of FOP was revealed. Mutant ALK2 was found to respond to Activin-A, whereas wild type ALK2 function is inhibited by Activin-A. The latter cytokine is induced locally upon tissue damage and inflammation. Moreover, therapeutic targeting of Activin-A was found to inhibit heterotopic ossification in a mutant ALK2 knock-in mouse model that is highly reminiscent to human FOP. This review will focus on these latest surprising findings and discuss the implication for treatment of FOP patients

Clustering and graph mining techniques for classification of complex structural variations in cancer genomes

    Get PDF
For many years, a major question in cancer genomics has been the identification of those variations that can have a functional role in cancer, and distinguish from the majority of genomic changes that have no functional consequences. This is particularly challenging when considering complex chromosomal rearrangements, often composed of multiple DNA breaks, resulting in difficulties in classifying and interpreting them functionally. Despite recent efforts towards classifying structural variants (SVs), more robust statistical frames are needed to better classify these variants and isolate those that derive from specific molecular mechanisms. We present a new statistical approach to analyze SVs patterns from 2392 tumor samples from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium and identify significant recurrence, which can inform relevant mechanisms involved in the biology of tumors. The method is based on recursive KDE clustering of 152,926 SVs, randomization methods, graph mining techniques and statistical measures. The proposed methodology was able not only to identify complex patterns across different cancer types but also to prove them as not random occurrences. Furthermore, a new class of pattern that was not previously described has been identified.Among others, this study has been supported by projects: SAF2017-89450-R (TransTumVar) and PID2020-119797RB-100 (BenchSV) from Science and Innovation Spanish Minstry. Hierarchical algorithms for causality retrieval in atrial fibrillation intracavitary electrograms

    Get PDF
Multi-channel intracavitary electrograms (EGMs), are acquired at the electrophysiology laboratory to guide radio frequency catheter ablation of patients suffering from atrial fibrillation (AF). These EGMs are used by cardiologists to determine candidate areas for ablation (e.g., areas corresponding to high dominant frequencies or complex fractionated electrograms). In this paper, we introduce two hierarchical algorithms to retrieve the causal interactions among these multiple EGMs. Both algorithms are based on Granger causality, but other causality measures can be easily incorporated. In both cases, they start by selecting a root node, but they differ on the way in which they explore the set of signals to determine their cause-effect relationships: either testing the full set of unexplored signals (GS-CaRe) or performing a local search only among the set of neighbor EGMs (LS-CaRe). The ensuing causal model provides important information about the propagation of the electrical signals inside the atria, uncovering wavefronts and activation patterns that can guide cardiologists towards candidate areas for catheter ablation. Numerical experiments, on both synthetic signals and annotated real-world signals, show the good performance of the two proposed approaches

Potential of Supercritical Carbon Dioxide Power Cycles to Reduce the Levelised Cost of Electricity of Contemporary Concentrated Solar Power Plants

    Get PDF
This paper provides an assessment of the expected Levelised Cost of Electricity enabled by Concentrated Solar Power plants based on Supercritical Carbon Dioxide (sCO2) technology. A global approach is presented, relying on previous results by the authors in order to ascertain whether these innovative power cycles have the potential to achieve the very low costs of electricity reported in the literature. From a previous thermodynamic analysis of sCO2 cycles, three layouts are shortlisted and their installation costs are compared prior to assessing the corresponding cost of electricity. Amongst them, the Transcritical layout is then discarded due to the virtually impossible implementation in locations with high ambient temperature. The remaining layouts, Allam and Partial Cooling are then modelled and their Levelised Cost of Electricity is calculated for a number of cases and two different locations in North America. Each case is characterised by a different dispatch control scheme and set of financial assumptions. A Concentrated Solar Power plant based on steam turbine technology is also added to the assessment for the sake of comparison. The analysis yields electricity costs varying in the range from 8 to over 11 g/kWh, which is near but definitely not below the 6 g/kWh target set forth by different administrations. Nevertheless, in spite of the results, a review of the conservative assumptions adopted in the analysis suggests that attaining costs substantially lower than this is very likely. In other words, the results presented in this paper can be taken as an upper limit of the economic performance attainable by Supercritical Carbon Dioxide in Concentrated Solar Power applications.Unión Europea (Programa Horizonte 2020) 81498

Influence of Working Fluid Composition on the Optimum Characteristics of Blended Supercritical Carbon Dioxide Cycles

Proceedings of ASME Turbo Expo 2021: Turbomachinery Technical Conference and Exposition GT2021 June 7-11, 2021, Virtual, OnlineThe supercritical Carbon Dioxide power cycle technology has attracted growing interest from the scientific community, becoming one of the most important options currently considered for CSP applications. This is thanks to its high thermal efficiency, even at moderate turbine inlet temperatures, and small footprint. Nevertheless, sCO2 power cycles require a fairly low compressor inlet temperature to exploit their full thermodynamic potential. When this cannot be achieved, as it is usually the case for Concentrated Solar Power plants where ambient temperatures are high, the interest of the technology is compromised. To compensate for this effect, the SCARABEUS project is working on the development of certain chemical dopants that could be added to the raw CO2, obtaining new working fluids with the same or even better performance than pure CO2 even at higher minimum cycle temperatures. This paper studies the impact of using CO2 mixtures blended with Hexaflurorobenzene (C6F6) and Titanium Tetrachloride (TiCl4). It is found that these mixtures enable thermal efficiencies that are higher than if pure CO2 were used. The efficiency gain can be as high as 3 percentage points, depending on the dopant used and the operating conditions considered. In addition to this absolute performance gain, the paper reveals that there are additional degrees of freedom that enable more effective cycle optimisation. These are the dopant molar content, not only its composition, and the cycle layout used. When this is studied, it is found that the optimum molar content ranges from 10 to 20% and that the layouts of interest when using mixtures are simpler than if plain CO2 were used. These results open the way for a significant performance enhancement of Concentrated Solar Power plants.Unión Europea, Horizon 2020, Nº81498
